Video: Summary of Recent Developments in Stem Cell and Regenerative Medicine from Kenneth Aldrich, Chairman of International Stem Cell Corporation


Greetings. This is Ken Aldrich, I am Chairman of International Stem Cell Corporation and thought I would share with you briefly some of the thoughts that I’ve had recently about developments in the stem cell and regenerative medicine area. One of the things that I have noticed from a lot of emails that we get, there is a fair amount of confusion out there about the significance of some of the new events that have taken place.

Specifically, I have gotten a lot of requests to explain what the impact of the recent announcement by Geron Corporation that they had entered FDA human trials might be. Well frankly, it is a very, very important step and one that benefits, I think, everyone in our industry and I wanted to comment on it a little bit. What that means is that one of the companies in our field has finally found the mechanism and found the procedures to begin the process of bringing cells to the clinic through human trials. So Geron, which is one of the largest companies, and has spent an enormous amount of money developing this, is now leading the path for all of us. I think we will learn from their experiences and it will make the path getting through to the FDA a lot more productive for all of us that follow.

There is another aspect to this however, that is unique to our company, International Stem Cell Corporation, in that we have also realized that the United States in only one part of the global market. And as a result, we’ve spent a lot of energy over the last year or so exploring foreign collaborations in those areas where perhaps the US is not the most attractive market. For example, we are working in India with replacing human corneas with corneas developed from our parthenogenic stem cells. The U.S. is probably not a major market for this because our systems here in this country allow for cornea transplants rather well. But in countries like India, as well as China and Korea and other places, the infrastructure doesn’t exist to harvest corneas from cadavers and deliver them and as a result, we have a wide open market there with enormous interest. I think that is one example of how the international market will impact the development of regenerative medicine.

We’re looking at that and we are looking at a variety of other areas and I’m sure other companies are doing the same. Eventually, we are all in this boat together to try to cure major diseases. We’re delighted with the progress with the FDA from companies here. We’ll be following in those footsteps when we can and we also be hopefully be leading the way in some of the international collaborations that may make all of us better off in the world of regenerative medicine.

International Stem Cell Corporation and The Automation Partnership Enter into Strategic Alliance to Automate Cornea Tissue Production

OCEANSIDE, CA – May 18, 2010 – International Stem Cell Corporation (ISCO.OB), www.intlstemcell.com, announced today that it had entered into a strategic alliance with The Automation Partnership (TAP), www.automationpartnership.com, to automate and scale up the production of stem cell-derived human corneal tissue. The alliance has been formed to create instrumentation for ISCO and its partners and affiliates to produce development and commercial volumes of donor tissue for cornea transplantation and to reduce the use of animals and animal eyes in safety testing of drugs, chemicals and consumer products.

Cornea-related loss or reduction of vision can be caused by physical injury, infections and degenerative diseases. In cases where cornea replacement is indicated, current medical practice typically involves a one-two hour outpatient procedure under local anesthesia using full or partial corneas from healthy human cadavers. 10 million people worldwide are candidates for such treatment, primarily in Asia and Europe where there is significant quantitative and qualitative shortage of human cornea donation.

Global efforts are underway to transition from the use of live animals and excised animal eyes to test drugs, chemicals and consumer products. For example, Europe’s Registration, Evaluation, Authorisation and Restriction of Chemicals (REACH) estimates a need to spend €270M and use 160,000 animals for eye safety testing alone to catch up with the back-log of insufficiently tested agents. In the US, the National Institutes of Health (NIH) and the Environmental Protection Agency (EPA) have launched a five-year program dedicated to finding new, non-animal technologies for toxicity testing of chemical compounds.

ISCO has discovered and filed for patents on a cell culture process for the synthesis of standardized, human, corneal tissue using stem cells. Histology, permeability and optical testing has demonstrated compatibility with natural corneas. Efforts are ongoing to further characterize this tissue and standardize and scale up its synthesis. Automation is necessary to produce sufficient, reproducible tissue for development and commercialization of the therapeutic and toxicity testing applications.

International Stem Cell Corporation Parthenogenetic Stem Cell Line Shown to Carry the Most Common Immune Genes Matched to the U.S. Population Retains Stem Cell Characteristics over Two Years in Culture, Which is Necessary for Therapeutic Value

OCEANSIDE, Calif. – March 2, 2010 – International Stem Cell Corporation (OTCBB: ISCO), www.internationalstemcell.com, announced today that human parthenogenetic stem cell lines (“hpSC”), created by scientists from its Research and Therapeutic Development Group, have demonstrated ability to maintain the essential properties of immortal stem cells throughout extended laboratory cultivation.

To date, ISCO has created 10 human parthenogenetic stem cell lines. One of these lines, named phSC-Hhom-4, has the most common set of HLA genes found in various U.S. population groups and may be useful to treat millions of people world-wide from a single cell line.

After more than one hundred passages over more than two years of continuous in vitro cultivation, the line phSC-Hhom-4 has maintained pluripotent stem cell properties and continues to be used in experiments to create therapeutic derivatives targeted for treatment of liver, cornea, retina and neurodegenerative diseases.

International Stem Cell Corporation Provides Strategic Update on the Company’s Cornea Transplantation Program

OCEANSIDE, CA – February 16, 2010 – International Stem Cell Corporation (ISCO.OB), www.intlstemcell.com, announced today that multiple international meetings taking place between ISCO’s executive management and researchers and clinicians from commercial entities in both Asia and Europe revealed significant clinical-commercial opportunities for the company’s stem cell-derived human cornea technology in those regions.

ISCO has discovered and filed for patents on a cell culture process for the synthesis of fully human, cornea-like structures using either its proprietary human parthenogenic stem cell (hpSC) technology or human embryonic stem cells. The structures are grown to clear hollow spheres with a size of 8-10 mm in diameter and contain tissues and cells similar to those found in normal human corneal tissue. Portions or all of these structures may be suitable for cornea transplantation in humans. Permeability and ocular histology testing has demonstrated compatibility with natural corneas. Future steps include scale-up of the manufacturing process and IND-enabling studies, to be conducted domestically and through international collaborations.

Cornea-related loss or reduction of vision can be caused by physical injuries, infections and a range of degenerative diseases that affect up to 10 million people worldwide. ISCO’s corneal structure may fit into existing medical applications where the surgical techniques are well established. Cornea transplantation has been refined greatly and is now typically performed as a one-two hour outpatient procedure using donated corneas from human cadavers. While most operations previously involved the entire corneal structure it is now common to selectively replace solely the damaged portion.

ISCO’s parthenogenic stem cell technology enables synthesis of corneal tissue that is immune matched for millions of people. This may significantly reduce the rejection rates of 15-30% experienced in current medical practice.

In the US 52,487 transplantations were performed in 2008. However, a shortage of corneal tissue has been a significant problem in much of the rest of the world. Only 3,000-4,000 procedures were performed annually in the UK and Germany due to limited cornea supply. In Asia, the shortage of corneas has been an even greater problem. For example, in Japan during a ten-year period only 16,000 transplantations were performed. China has had over 2 million patients on waiting lists yet only a few thousand procedures have been performed annually. Over 3 million Indians are reported to be blind due to corneal defects.

Dr Radhika Tandon, Professor of Ophthalmology and Officer-in-charge at the National Eye Bank, All India Institute of Medical Sciences (AIIMS) in New Delhi says: “Corneal vision impairment is a large medical problem in India and other developing countries. India has access to less than 20,000 suitable corneas per year yet would need 200,000 corneas to take care of the existing backlog and the new cases added each year. Supply of synthetic human corneas would alleviate the problem and provide great socio-economic benefit by enabling millions of Indians to get back to work and live a more normal life.”

International Stem Cell Corporation and Reproductive Medicine Associates of New York Announce Breakthrough in Creating an Abundant and Ethical New Source of Human Pluripotent Stem Cells

Oceanside, Calif., October 27, 2009 International Stem Cell Corporation (OTCBB:ISCO, www.internationalstemcell.com) and Reproductive Medicine Associates of New York (RMA of New York) have shown that immature, unfertilized human eggs can be used to create “parthenotes” from which human parthenogenetic stem cell lines may be derived. Immature eggs are normally discarded during standard in vitro fertilization (IVF) procedures due to their inability to create an embryo. This discovery vastly increases the opportunity for ISCO to create new stem cell lines and hastens the time when “banks” of stem cells that match a patient’s immune system may be available. This vision of a “patient matched stem cell bank” could function similarly to how blood banks currently store and distribute blood products.

A “parthenote” results when an egg divides without using sperm (no fertilization) and can yield “parthenogenetic” stem cells that have similar medical potential as embryonic stem cells to create various human tissues for therapy yet have the additional medical advantage of allowing immune-matching to reduce the chance of rejection - a significant potential problem with the use of embryonic stem cells. Parthenogenesis also addresses critical ethical issues by eliminating the need to use fertilized embryos.

According to Dr. Alan Copperman, Director of the Division of Reproductive Endocrinology and Infertility and Vice-Chairman of the Department of Obstetrics, Gynecology, and Reproductive Science at Mount Sinai Medical Center in New York, “This scientific breakthrough opens many new opportunities for our clinical and scientific teams to explore. Transforming immature oocytes into stem cell lines capable of differentiating into a variety of cell types has the potential to revolutionize the field of regenerative medicine.”

Dr. Copperman is an accomplished infertility specialist who practices at Reproductive Medicine Associates of New York and is a member of ISCO’s Scientific Advisory Board.

This ground-breaking work was presented at the American Society for Reproductive Medicine’s 65th Annual Meeting held in Atlanta, GA on October 19, 2009.

Jason Barritt, PhD, Scientific Director of RMA of New York noted, “This work shows that immature oocytes that are normally unusable and discarded in standard IVF procedures may be used for scientific innovation and clinical application. International Stem Cell Corporation and Absorption Systems Announce Potential Stem Cell Alternative to Live Animal Testing for Corneal Damage


International Stem Cell Corporation (OTCBB:ISCO)(www.internationalstemcell.com), the first company to perfect a method of creating human "parthenogenetic" stem cells from unfertilized eggs, announces positive results of its collaboration with Absorption Systems to study ISCO's stem-cell-derived human corneal tissue as an alternative to live animals for drug testing. Initial results using non-animal or "in vitro" tests indicate an excellent correlation between the rates at which drugs pass through ISCO's lab-grown corneal tissue and rabbit corneal tissue. The results offer great promise for reducing the use of living animals for eye safety testing. ISCO's human corneal tissue is created from parthenogenetic stem cells in the laboratories of Lifeline Cell Technology (Walkersville, MD), ISCO's wholly-owned subsidiary.

According to Dr. Chris Bode of Absorption Systems. "The rank order correlation of the eight drugs tested so far is almost perfect."

The collaboration between the companies is focused on using Absorption Systems' know-how in creating assay systems to develop superior preclinical methods of testing drugs. Such methods can also reduce the use of laboratory animals currently necessary for other tests, including the very large global commercial need for a reliable and reproducible non-animal method for eye safety testing of consumer products.

In the past, researchers have grown corneal cells in the laboratory, but ISCO is believed to be the only company to grow a self-assembling corneal construct from parthenogenetic stem cells that is suitable for toxicity testing and possibly suitable for corneal transplants. ISCO's corneal constructs include cell layers and structures normally found in a human cornea, as confirmed by an independent third-party laboratory. ISCO's proprietary lines of parthenogenetic stem cells remove the need for fertilized embryos and, in a therapeutic setting, may minimize the threat of immune rejection. This breakthrough was published in the peer-reviewed journal Cloning & Stem Cells (Volume 9, Number 3; September 2007:432-449 and, Volume 10, Number 1; March 2008: 11-24).

Kenneth Aldrich, CEO of International Stem Cell Corporation, Addresses Some Questions Regarding Letter in Response to NIH Proposed Guidelines

In light of recent comments made by International Stem Cell in response to the NIH proposed Guidelines, I have been asked a lot of questions. Let me try to answer a few of the most common ones.

Q: Will the proposed guidelines hurt International Stem Cell unless they are revised?

A: No. They will be an inconvenience and perhaps force us to accept funding from foreign instead of US sources, but will not change the fundamental prospects of our company at all. We have been operating under the same kind of restrictions since the founding of our company and can continue to do so. NIH funding is of far more importance to academic researchers than to companies and a change specifically permitting Federal funding for Parthenogenesis research would enable more researchers in the US to use our cells. That would enable us perhaps to do more of our work here at home, but we truly are an international company and expect to be able to obtain the kind of funded research and researchers even if the proposed rules are not changed. However, we would greatly prefer to develop our technology with US based scientists in US universities if possible.

Q: Why then are you so opposed to the present proposed Guidelines?

A: The present proposals specifically exclude the use of Parthenogenesis (our technology) and SCNT (technology that we have licensed but do not currently use). Both of these are potentially very valuable sources of cells for the treatment of human diseases and can do things that can't be done with Embryonic stem cells or the iPS cells that are derived from genetically re-programmed human tissue. For example, parthenogenetic stem cells can be matched to the immune systems of millions of individuals, thus allowing the efficient creation of true stem cell banks for the population. Different stem cell technologies increase the potential for new medical breakthroughs and we believe that parthenogenesis is the strongest technology with the most medical potential. If the US government restricts the kind of technology it will support, whether because of politics or ignorance of the facts, it could deprive millions of people of the benefit of cures that might otherwise have been discovered.

Q: Why do you think the NIH has restricted its funding in these Guidelines?

A: I believe there are several issues, but none of them apply for Parthenogenesis. The NIH does not want federal money spent to derive human embryos for research use only. This does not apply for Parthenogenesis, because Parthenogenesis requires unfertilized human eggs – not fertilized human embryos. We believe this places Parthenogenesis on higher ethical ground. In addition, all of our Parthenogenetic lines have been created by women who entered IVF clinics with the intent of undergoing in-vitro fertilization (IVF), not the intent of creating stem cell lines.

I can only speculate, but strongly suspect that an additional reason may be a fear that somehow these technologies will lead to the need for thousands of human eggs and that women donors will be abused or taken advantage of to obtain such eggs for research. Nothing could be farther from the truth. The reality is that Parthenogenetic stem cells are derived from the excess eggs that fertilization clinics will eventually throw away. The only relevant difference between how stem cells are created through Parthenogenesis and how embryonic stem cells are created is whether or not the eggs are fertilized before being used as a source of stem cells. That has no impact on the donor at all. She produces the same number of eggs in the same way and only the excess are used for research, all under the same kinds of consent regulations. In addition, Parthenogenesis is an efficient process and does not require large numbers of eggs to produce a stem cell line. In fact, because Parthenogenesis can create a single line from a single donor that can be immune-matched to millions of individuals from multiple racial groups, once a bank of these lines is created, the need to obtain more human eggs will be greatly diminished or eliminated.

I hope these comments will clear up some of the mystery.

International Stem Cell Corporation Receives Third $1M Tranche under a $5 Million Financing

International Stem Cell Corporation (OTCBB:ISCO) has received the third $1 million tranche of an anticipated private equity financing of up to $5 million to be funded over the next several months. The total amount of the financing will allow the Company to move forward with the construction of its new cGMP cell culture facility and to continue its therapeutic research, including ongoing pre-clinical trials. The money will also be allocated to fund equipment, product development and marketing requirements to increase revenues in the Company's subsidiary, Lifeline Cell Technology, which makes and sells specialty cells and growth media.

ISCO Enters into an Agreement with Cytograft Tissue Engineering to Develop a Therapeutic Medium for Cytograft's Living Vascular Grafts

Living Laboratory Grown Blood Vessels from the Patient's Own Cells

International Stem Cell Corporation (OTCBB:ISCO) (www.intlstemcell.com), the first company to perfect a method of creating human "parthenogenetic" stem cells, which are derived from unfertilized human eggs, is working with tissue engineers at Cytograft Tissue Engineering, (Novato, CA), to design a custom cell culture medium to cultivate living human vascular grafts; grafts which are expected to be used in future human clinical trials.

Cytograft's living human product, called Lifeline™ (trade-name not associated with Lifeline Cell Technology), is a promising technology that will be used as a coronary graft for coronary bypass procedures, as a peripheral graft to prevent lower limb amputations and as a living tissue shunt for hemodialysis patients.

The new medium will be developed and produced by scientists at ISCO's wholly-owned subsidiary, Lifeline Cell Technology®, a producer of human cells and media products.

Article Details ISCO's Solutions To Ethical And Immune Rejection Issues

International Stem Cell Corporation (OTCBB: ISCO) (www.internationalstemcell.com), the first company to perfect a method of creating human "parthenogenetic" stem cells from unfertilized eggs, has been featured in the January 31, 2009, edition of The Economist, the authoritative weekly publication focusing on international politics and business news and opinion.

The article, titled "Embryonic Stem Cells: Can I Serve You Now?," details how ISCO's unique parthenogenetic approach helps resolve two of the conundrums that have long shadowed stem cell research: ethical issues and immune rejection difficulties. "ISCO has found a way of producing embryonic-like stem cells from unfertilized eggs. The egg is chemically stimulated to create a group of cells that form a non-viable (and unfertilized) 'embryo' . . . something that could not be implanted into a woman's womb and produce a child. Nonetheless, the cells it contains have the same characteristics as stem cells. Besides the ethical advantages this procedure has, it could also have medical ones. Because lines of stem cells created in this way have only one parent, they are immunologically simpler than normal embryonic cells—in other words they have a smaller variety of the proteins that trigger immune rejection."
