International Stem Cell Corp. Progressing Despite Incompetent MSM

Dear Breakthrough Technology Alert Reader,
"American journalism (like the journalism of any other country) is predominantly paltry and worthless. Its pretensions are enormous, but its achievements are insignificant."

Last week, an article by Rob Stein appeared in The Washington Post titled "Scientists Report Possibly Crucial Advance in Human Embryonic Stem Cell Research." Stein, whose credentials include science editor positions at NPR and The Washington Post, describes the use of cloning techniques to turn oocytes, immature eggs, into embryonic cells. The first one-sentence paragraph is as follows:
"Scientists reported Wednesday that for the first time, they had used cloning techniques to generate embryonic stem cells containing the genes of specific patients."

Frankly, I don't know what that sentence means. Somatic cloning of stem cells has been going on for years at places such as the Harvard Stem Cell Institute and the University of California San Francisco. It's not a particularly difficult thing to transfer adult DNA into an embryonic stem cell. Maybe the researchers said that it was the first time that "they" had used cloning techniques to make ESCs and the writer misinterpreted their statements. I have no idea.

Moreover, that sentence includes the word "patients." There were no patients involved in the study. It was a laboratory demonstration of a cloning technique in which DNA from mature cells is transferred into immature eggs, oocytes. Then, the oocytes transformed into stem cells with the implanted genes. The interesting thing about the procedure, which you can access here, is that the DNA from a normal cell, from two parents, was added to a cell with only a mother's genes.

This resulted in a cell with the DNA of three people -- a triploid cell. While fascinating, it also raises all kinds of scientific and safety issues. We have no idea what triploid cells would actually do if they were implanted in a patient. With extra haploid matching points, they might even be immunocompatible with no one.

I do, however, have a great script idea if somebody with money would like to hear the pitch. I'm thinking Species meets Mommie Dearest. Regardless, the possibility that triploid cells will be therapeutically useful in the foreseeable future is very low.

This brings me to the real flaw in the story. Read the second sentence/paragraph:
"The step marks a possibly pivotal advance toward the long-sought goal of creating stem cells that could be used to treat many major diseases, because they would not be rejected by patients' immune systems."

Balderdash. There are at least two solutions already available for dealing with immune rejection -- induced pluripotent stem cells (iPSCs) and human parthenogenic stem cells (hpSCs).

The iPS cell technology uses an individual's own cells to create cells that are functionally equivalent to embryonic stem cells. These would be compatible with the donor. The other solution is the human parthenogenic stem cells developed by International Stem Cell Corp. (OTCBB:ISCO). These, scientists believe, will solve the immune rejection problem on a mass-market basis.

ISCO long ago demonstrated that they have made the "pivotal advance" using oocytes, to which the Post article refers. ISCO scientists discovered and patented the techniques that successfully create human parthenogenic stem cells (hpSCs), which can be programmed to become any other cell type. With ISCO's cell banks, which will match the various HLA profiles of the human species, the immune rejection problem will be solved for the vast majority of the human race.

You will recall from my recent issue about ISCO that the company has already begun collecting oocytes to create these parthenogenic cells lines. It does so with complete approval of regulatory and ethical agencies.

As you know, it took ISCO many years and millions of dollars to accomplish their technological breakthrough. Interestingly, it had previously been done, apparently in an accidental and nonreproducible manner. Stem cell scientist Woo Suk Hwang of Seoul University announced in 2004 that he had produced a stem cell line derived from an embryonic human clone. Later, it was discovered that Hwang did not have a cloned stem cell line. He had a parthenogenic stem cell line.

Though Hwang was disgraced, the irony is that cloned stem cell lines are far easier to produce than hpSC lines. I don't understand all the details about what he did, wrong or right. The irony is this, however: If he had presented his stem cells as hpSCs, he might have been acclaimed a scientific hero, instead of the super-villain he is considered today.

The Washington Post article goes on to cite the usual stem cell controversies. Those opposed to ESC research have voiced opposition to the triploid stem cell technology when asked. They believe the triploid cells are embryonic stem cells. Since they are, in a sense, fertilized, that may be true. It's not at all clear, though, that they could develop into an embryo. I doubt it.

Also, the fact that some fertility therapy patients were paid well for oocytes unneeded in the fertilization process has caused serious concern among feminist commentators. They are seriously concerned. Seriously.

The big point, however, is that this article badly misrepresents the state of the science. ISCO scientists have long had the ability to create pluripotent stem cells using oocytes. More to the point, cells made using their process have none of the ethical issues of these triploid cells. They are not fertilized and cannot become embryos. They require no genetic manipulation. No foreign DNA is inserted into them. The company does not pay for oocytes and has followed every ethical guideline acquiring them.

The Wall Street Journal apparently picked up on the "breakthrough" story and helped spread the disinformation, as did other news outlets and bloggers. I'm not, per se, a journalist, but if I were writing for a nationally read publication that purports to present science accurately, I would have started by checking around to see if there were other perspectives on stem cells from parthenogenic oocytes.

In fact, I did a simple Google search using key words "parthenogenic" (or "parthenogenetic"), "stem" and "cell." The first page yielded this link to ISCO's description of their human technology.

Then, looking around the site, we find a list of respected scientific partners who are assisting in further research on ISCO's hpSCs, as well as some of their research areas.

They include:

Novocell Inc., San Diego, Calif., endocrine pancreas cells.
University of California, San Francisco, Calif., hepatocyte cells.
University of California, Irvine, Calif., retinal pigment epithelium.
University of Wurzburg, Germany, neuronal cells.
The Scripps Research Institute, La Jolla, Calif., characterization of human parthenogenetic stem cells.

None of these organizations is inaccessible. You can find their phone numbers on the innerwebz, and it has been my experience that scientists are more than happy to tell journalists what's really going on. Oh, well.

Having spent so much time in debunking mode, now I feel like I ought to at least bring you up-to-date on some new developments at ISCO. Several important events have, in fact, occurred lately.

ISCO to Expand Cosmeceutical Product Lines

One, ISCO has been perfecting the process of growing and preserving the active ingredient in parthenogenic stem cells used for their cosmeceutical skin care product line. As a result, they've been able to stockpile enough of the growth factors found in stem cells to allow additional marketing efforts. In the past, availability of product has limited those efforts.

Moreover, the company has announced development of several new skin care products, including one designed to work on the sunken area under the eyes. The skin care line, while not cosmically meaningful, is doing very well for the company and is on track to fund serious work, such as ISCO's liver cell science.

Also, ISCO has brought in an experienced corporate executive to help move the company forward. Dr. Andrey Semechkin, currently CEO, will become co-chairman. Kurt May, currently senior vice president, will take the helm as president and COO.

California Health Institute Interviews Jeffrey Janus - CEO of Lifeline Cell Technology


Jeffrey Janus serves as director and senior vice president of operations of new CHI member International Stem Cell Corp. and president and chief executive officer of Lifeline Cell Technology, one of the company’s subsidiaries. International Stem Cell Corp. (ISCO.OB) is a publicly traded stem cell therapy company with research and manufacturing facilities in Oceanside, Calif., and Walkersville, Md. The company’s technology revolves around its discovery of a proprietary and unique class of stem cells called human parthenogenetic stem cells (hpSC). These cells have distinct medical, practical and ethical advantages over embryonic and adult stem cells. They allow immune-matched stem cells and therapeutic cells to be “banked” and available immediately for millions of patients who are in critical need and cannot wait to derive cells from their own bodies. In addition to Lifeline Cell Technology, the company has another subsidiary called Lifeline Skin Care.

Janus is trained in biochemistry and business management and has more than 20 years experience focused on cell-based businesses. He is a member of the team that discovered parthenogenesis and is published in the stem cell field. After joining International Stem Cell Corp., (ISCO) Janus subsequently founded Lifeline Cell Technology to meet a growing need for media and human cells in pharmaceutical drug screening, consumer product testing and basic research at universities and government laboratories and to provide revenue and operational infrastructure for ISCO. The CHI Blog recently caught up with Janus to find out the latest on the company.

Q: How did your company get started? A: We started this company based on the work of Elena Revazova, M.D., Ph.D., a scientist well known in Russia who had a dream of curing diabetes using embryonic stem cells. She came to the United States to work and her talent and expertise in growing human cells was discovered by ISCO’s founders, who decided to form a company around her knowledge and skill. At the time, U.S. President [George W.] Bush was restricting the use of embryonic stem cells on ethical grounds, and there were also patent issues around embryonic stem cells, as there still are. We recognized that the ethical issue was important, but medially the most important problem with stem cell therapy was likely to be immune rejection. We realized we could address these issues by developing the technology called parthenogenesis and mitigate delays from funding and restrictions by working in Russia. So Dr. Revazova went back to Russia, and we set up a collaboration in Moscow to begin her work with parthenogenesis. Today our company has all of the intellectual property rights to parthenogenesis, a very powerful technology. We have also recently brought in Andrey Semechkin, Ph.D. as our CEO. Dr. Semechkin is a well-known scientist in the field of systems analysis and an accomplished businessman.

Q: How does parthenogenesis work? A: It’s the derivation of stem cells from an unfertilized human egg. The ethical issue surrounding work with embryonic stem cells is caused by the fact that embryonic stem cells are derived from a fertilized embryo, which has the potential to be a human being. However, if you do not fertilize the egg and yet you can derive stem cells from it that are functional, you’re not destroying a viable human embryo—and that’s exactly what Dr. Revazova did. We perfected parthenogenesis and brought it back to the United States. As a result, we have been able to overcome the ethical issue surrounding using embryonic stem cells with parthenogenesis.

Q: What are your technology’s other advantages? A: Parthenogenesis makes embryonic stem cells (or what we call parthenogenetic stem cells) that can be immune matched to millions of people. Using embryonic stem cells, the way they are currently made, is sort of like trying to do a bone marrow transplant between one person and another picked at random without making sure you have a match. If someone needs to have a bone marrow transplant, they usually go to brothers or sisters first and try to do an immune match. For a different set of reasons a similar situation exists with blood transfusions, although type O blood can be given to almost everyone. Our cells are similar in that the parthenogenic stem cells can be immune matched to many people, and that’s the unique quality of our cells.

Q: What are the biggest opportunities for your business going forward? A: We are creating a bank of hpSC that are “pluripotent” and carry common immune types that will match a large percent of the U.S. population, and this is a huge opportunity. These will be clinical grade and will be made in our new manufacturing facility located in Oceanside, Calif. Our biggest opportunity is the potential ability of our stem cells to be universally utilized for therapy. Scientists across the world are working on embryonic stem cells and figuring out ways to make therapeutic cells such as liver cells or nerve cells for a whole host of diseases. Eventually these therapies will need a cell or process that will minimize immune rejection. Our cells can be immune matched to millions of persons and are thus a solution for this need. So in a way, much of the work that’s going on right now across the world with embryonic stem cells accrues to our benefit. In addition, we are focused in four distinct areas—diabetes, liver disease, retinal and corneal disease, and nerve disease. We are currently growing cells to cure corneal blindness and have actually grown cornea tissue. We’re working with the University of California, Irvine to grow cells with a retina for macular degeneration. We have grown cells that are very similar to liver cells that are also related to a cell type called beta cells, which may be useful for diabetes. Collaborations with companies and universities present strong opportunities, and we’ve collaborated with Novocell in San Diego to further our work with diabetes, and we’re collaborating with UC San Francisco to test our liver cells derived from our parthenogenic stem cells and with researchers in Germany to study nerve cells generated from our stem cells.

Q: Tell us a little bit about your subsidiaries. A: One unique thing about our company is that we are a research-oriented biotech company that actually has income. One of our subsidiaries, Lifeline Cell Technology, is growing very nicely (with a 150 percent increase in sales over the last year) by selling research products to grow human cells and study human disease. Lifeline has more than 70 products and will be releasing more than a dozen more in 2010. Lifeline Skin Care was created in 2009 based on our discovery that derivatives from our parthenogenetic stem cell technology have proven to be beneficial to human skin. Lifeline Skin Care is developing several products and is beginning early-stage clinical trials with these skin products. We anticipate that these skin care products will help to generate income and fund our continuing stem cell therapeutic research.

Q: What are your company’s greatest accomplishments so far? A. We have successfully created 10 human parthenogenetic stem cell lines, one which carries the most common immune type in the United States and matches over a hundred million persons across the world. We are a fast-growing company with more than 12 scientists working in various areas of therapy and product development. Our stem cells have proven to be able to create cells that may be useful in therapy, including liver-like cells, corneal cells, retinal cells, nerve cells and cell types that may ultimately be useful in the treatment of diabetes. We have set up collaborations with major universities and researchers across the world. The amazing thing about our company is that we have developed into a company that has manufacturing, products, sales, quality control, therapeutic research, and an accounting department in such a short time. We have all the workings of a fully functional product manufacturing and therapeutic research company. It amazes me that we are making sales, whereas most companies our size are basic research and development companies. We know how to make human cells and freeze, store and manipulate them so that they are clinical grade. I think our technology, our knowledge of cell culture and our ability to manufacture are three very strong reasons that we have been successful.

International Stem Cell Corporation Opens Facility for Development and Production of Clinical-Grade Stem Cell Products

OCEANSIDE, CA – February 4, 2010 – International Stem Cell Corporation (ISCO.OB), www.internationalstemcell.com, announced today that its new cell production facility has passed final building inspection, which will enable development and manufacturing of the company and its partners’ clinical-grade stem cell products.

ISCO has designed and built a unique cell manufacturing facility in Oceanside, California consisting of separate “suites” for development and production of different cell types from the company’s proprietary human parthenogenic stem cell (hpSC) technology. The facility is located in close proximity to the fertility clinics that provide donated human eggs (oocytes) under ISCO’s recently-established partnerships and also near leading Californian research institutions with whom ISCO collaborates on fundamental stem cell biology and therapeutic applications.

ISCO will implement its parthenogenic stem cell processes at this facility during 2010 and adopt current good manufacturing practice (cGMP) standards.In parallel, ISCO will collaborate with world-leading scientists to demonstrate the therapeutic applicability and the potential immune-rejection advantages of hpSC lines relative to other stem cell classes.

Like embryonic stem cells (ESCs), hpSCs are pluripotent (i.e. have the capacity to become almost any cell type in the body), yet avoid ethical issues associated with destruction or use of viable human embryos. Unlike induced pluripotent stem cells (iPSs), hpSCs do not require manipulation of gene expression back to a less differentiated stage, which may prove to be a safety or regulatory obstacle. However, unlike both ESCs and iPSs, hpSCs can be created in a homozygous form such that each line can be an immunological match for millions of patients.

International Stem Cell Corporation Receives Third $1M Tranche under a $5 Million Financing

International Stem Cell Corporation (OTCBB:ISCO) has received the third $1 million tranche of an anticipated private equity financing of up to $5 million to be funded over the next several months. The total amount of the financing will allow the Company to move forward with the construction of its new cGMP cell culture facility and to continue its therapeutic research, including ongoing pre-clinical trials. The money will also be allocated to fund equipment, product development and marketing requirements to increase revenues in the Company's subsidiary, Lifeline Cell Technology, which makes and sells specialty cells and growth media.

International Stem Cell Corporation Applauds President Obama's Decision to Allow Funding of Parthenogenetic Stem Cell Research

International Stem Cell Corporation (OTCBB:ISCO) (www.intlstemcell.com), the first company to perfect a method of creating human stem cells from unfertilized eggs, applauds President Obama's signing of an executive order to allow funding of stem cell research. ISCO's unique human stem cells, called "parthenogenetic" stem cells, have ethical advantages since they do not harm a viable human embryo and have medical advantages since they can be immune matched to hundreds of millions of people across the world.

Monday, in an East Room ceremony at the White House, President Obama signed an executive order to lift the Bush administration's restrictions on federally financed human embryonic stem cell research. This decision provides Congress with an opportunity to overturn a thirteen-year-old law, called the Dickey-Wicker amendment, which specifically bans the use of tax dollars for research in which human embryos are destroyed, discarded, or knowingly subjected to risk of injury.

"Tissue-specific differentiated cells from human stem cells open up the possibility of cell-based therapy for untreatable degenerative and heredity diseases. The greatest risk posed with these cells for transplantation is that of immune rejection. Matching donor and recipient for HLA antigens is necessary for transplant survival. ISCO already has a human parthenogenetic 'HLA homozygous' stem cell line that carries the HLA type found most commonly within the US population. Tissue-specific differentiated cells from this line can be used for cell-based therapies of hundreds of millions of patients of different ages, races and sexes," says Elena Revazova, M.D., Ph.D., Chief Scientific Officer of ISCO.

Recently, the company reported that by using its proprietary technique, cells from a single donor could be immune matched to hundreds of millions of patients.

Since a single line of these cells may reduce immune rejection issues in large segments of the population, parthenogenetic stem cells could be enormously valuable as a treatment of choice for diseases including liver disease, heart disease, and macular degeneration, and of particular value in treating diseases with hereditary links such as diabetes, Parkinson's and Alzheimer's diseases, hemophilia, retinitis pigmentosa and many others in which cells from the patient's own body cannot be used for therapy because the patient's cells carry a corresponding genetic defect. For these patients it will likely be necessary to use HLA matched human parthenogenetic stem cell lines derived from healthy donors. International Stem Cell Corporation Cuts a Deal with BioTime, Inc., and Embryome Sciences, Inc., to Provide Unique Human Stem Cell Lines for Research

International Stem Cell Corporation (OTCBB:ISCO), through its wholly owned subsidiary Lifeline Cell Technology (Lifeline), has signed a manufacturing and distribution agreement with BioTime, Inc., (OTCBB:BTIM) (Alameda, CA) and BioTime’s wholly owned subsidiary Embryome Sciences, Inc., to jointly produce and distribute hundreds of new standardized human and animal stem cell lines, along with corresponding data and reagents. The mutual goal is to provide the “picks and shovels” for scientists mining the stem cell field for therapeutics in the emerging fields of regenerative medicine and pharmaceutical drug discovery.

These unique stem cell lines (called “progenitor” cell lines) possess the potential to expand and become specific types of tissues and cells such as heart, liver, skeletal muscle, bone, retinal, nerve, pancreas and others – all necessary to study various human diseases where regenerative medicine shows great promise.

“There is a large opportunity in providing high-quality progenitor cell lines and other basic research tools for use in stem cell research and drug discovery, especially since $3 billion in research funds for California’s Proposition 71 are now becoming available. Under the collaborative production and manufacturing agreement, the parties intend to manufacture EspyTM cell lines (derivatives of hES cells that send beacons of light in response to the activation of particular genes), as well as a host of supplies scientists will utilize in the field of stem cell research. The progenitor cell lines will be produced and distributed in joint efforts utilizing Embryome Sciences’ proprietary “Embryomics ” technology, International Stem Cell’s proprietary parthenogenetic stem cell lines derived from unfertilized human eggs and technology and approved hES cell lines licensed from the Wisconsin Alumni Research Foundation (WARF). Data on these lines will be presented on Embryome Sciences’ future Embryome.com online database. International Stem Cell will contribute its manufacturing and quality control expertise backed by a staff with over 150 years of experience in the field.

Summing Up Stem Cells by Kenneth Aldrich

As I have been traveling around the U.S. talking to editors and financial analysts about our company, International Stem Cell Corporation (“ISCO.OB), I am constantly struck by how much confusion there is about what stem cells are, what the difference is between “adult” and “embryonic” stem cells, and how they can be used in developing ways to treat dreadful diseases. Let me offer some thoughts that may be helpful.

First of all, let me refer you to what I think is the finest explanation of stem clls for the non-scientist I have ever read. It is contained in an article by Dr. Douglas Melton of Harvard University that was recently published in the Bulletin of the American Academy. (http://www.amacad.org/publications/bulletin/summer2007/stemcell.pdf). I urge anyone interested in understanding the world of stem cells to download it and read it.

However, there is one thing not covered by the article because it was not widely known at the time. Dr. Melton speaks of the differences between adult stem cells, which are limited in their capacity to become other types of cells, and embryonic stem cells, which can become any cell in the body. Embryonic stem cells are also at the heart of all the current debate about the ethics of stem cell research because they are obtained from fertilized human eggs.

What was not widely known at the time of Dr. Melton’s article is that there is a way of creating stem cells that have the same capacity to be changed into any cell in the body that might be needed for therapy or research, but do not use fertilized eggs. These are stem cells developed by my company, International Stem Cell Corporation, created by a process called Parthenogenesis. The cells are called Parthenotes. At the risk of sounding like a commercial, let me explain why we think these cells are so important.

The first reason is obvious. By avoiding the need to use fertilized eggs, we believe we can put to rest the fears of those who believe that destroying a fertilized embryo is destroying a human life. For reasons that can get quite technical, it is generally agreed that in humans a Parthenote cannot be implanted back into the donor’s womb and grow to be a human being. It is a cluster of cells that can still produce any cell in the body, but lacks a critical element necessary to allow it to attach to the uterus and become a baby.

The second reason is even more important. Any implanting of a cell to treat a disease is like transplanting a miniature organ. If you don’t have a match with the patient’s immune system, the body will reject the cell. It is one of the big problems not yet solved in cell transplant therapy and millions of dollars have been spent trying to find ways to trick the body into not attacking the implanted cell.

We think we have a better way by far. Because Parthenotes contain only the dna of the mother from whose egg they were derived, and not that derived from a father’s sperm, it is possible to create cells from a single stem cell line that can match the immune profile of hundreds of millions of people. That means that instead of having to search through hundreds of thousands of cell lines trying to find one that matches, a very small number of lines might suffice for most of the world’s population.

For the first time in its three-year existence, the state taxpayer-funded stem cell institute is offering grant money to biotechnology companies.

Ten companies, including at least two in San Diego, said they plan to apply for money for so-called disease teams, a cutting-edge concept of bringing academia and industry together to share expertise and perhaps speed the lengthy and expensive process of getting new therapies and diagnostics to market.
